一般描述
在有氧呼吸期间氧气减少以及在细胞内各种酶体系的作用下,可生成活性氧类物质(ROS)。在生理水平上,ROS有助于细胞信号传导和宿主防御能力。当ROS生成增加至超过生物系统的解毒能力时,将导致氧化应激和细胞损伤。对细胞的主要损伤来源于ROS诱导的大分子改变,如膜脂质的多不饱和脂肪酸、必需蛋白质和DNA。已有研究证明ROS与疾病状态有关,如阿尔茨海默病、帕金森病、癌症和衰老。
应用
本试剂盒适于通过酶标仪、流式细胞仪或带TRITC滤镜的荧光显微镜检测细胞内的ROS。
细胞内活性氧类物质(ROS)检测试剂盒(荧光法)已可用于评估机械通气(MV)对线粒体的损害。它还被用于测量细胞内ROS的产生。
生化/生理作用
细胞内ROS检测试剂盒(荧光法)是一种灵敏的一步式荧光测定法,仅需1小时孵育即可检测活细胞中的细胞内ROS(特别是超氧化物和羟基自由基)。ROS与细胞可透过性传感器反应,产生的荧光产物(λex = 540/λem = 570 nm)与ROS含量成比例。该测定可使用方便的96孔或384孔微量滴定板。
