凝胶过滤层析是一种确定蛋白质大小和分子量的方法。分馏是基于分子在树脂孔中的扩散。较大的蛋白质不易进入树脂孔，但比较小蛋白质更快地通过柱液体积。这些蛋白质分子按分子量递减的顺序从柱中洗脱。
应用
凝胶过滤标记物可用于蛋白色谱和凝胶过滤色谱。凝胶过滤标记物已用于研究糖皮质激素诱导的淋巴细胞溶解作为免疫系统内细胞凋亡的模型系统。凝胶过滤标记物也已用于分离玉竹凝集素，其对Vero细胞具有显著的抗HSV-II活性，对人黑素瘤A375细胞具有细胞毒性并可以半胱天冬酶依赖性方式诱导细胞凋亡。
适用于蛋白质分子量 6,500-66,000 Da的凝胶过滤标记物试剂盒已用于尺寸排除色谱法中的标准品。
