General description
Phosphatase of regenerating liver (PRL-3), also known as protein tyrosine phosphatase type IVA, member 3 (PTP4A3) is a dual-specificity phosphatase. It belongs to the phosphatases of regenerating liver family. The PTP4A3 gene is localized on human chromosome 8q24.3.
Immunogen
Synthetic peptide corresponding to amino acid residues of human PRL-3 with N-terminal added cysteine, conjugated to KLH. The corresponding sequence is identical in mouse.
Application
Anti-PRL-3 antibody produced in rabbit has been used in western blotting and immunohistochemistry.
Biochem/physiol Actions
Anti-PRL-3 recognizes human phosphatase of regenerating live (PRL)-3 and does not cross-react with human PRL-1 and PRL-2.
Phosphatase of regenerating liver (PRL-3) promotes epithelial-mesenchymal transition. It also plays an essential role in cancer cell migration, invasion and metastasis. It dephosphorylates serine, threonine and tyrosine residues. The protein is overexpressed in various cancers. PTP4A3 associates with transcription factors and regulates gene transcription. In addition, it also regulates breast cancer cell proliferation and invasion. Thus, this protein acts as a potent therapeutic target for breast cancer.
Physical form
Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.
Preparation Note
For continuous use, store at 2-8 °C for up to one month. For extended storage, freeze in working aliquots. Repeated freezing and thawing is not recommended. Storage in frost-free freezers is also not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilutions should be discarded if not used within 12 hours.
