生化/生理作用

吡菲他林-α是 p53 介导的细胞凋亡和 p53 依赖性基因转录如细胞周期蛋白 G、p21/waf1 和 mdm2 表达的可逆性抑制剂。吡菲他林-α在遗传毒性应激(如 UV 照射)和细胞毒性化合物(包括多柔比星、依托泊苷、紫杉醇和胞嘧啶-β-D-阿拉伯呋喃糖苷)处理后增强细胞存活。吡菲他林-α保护小鼠免受致死性全身 γ-辐射,且不增加癌症发病率。这种保护作用在 p53 缺失的小鼠或 p53 基因显性负突变体表达的细胞中没有观察到。p53 缺陷细胞系中 p53 的瞬时表达赋予保护作用。
