General description
天然完整的人纤溶酶原是791个氨基酸的糖蛋白，具有多达 24个二硫键。纤溶酶原包含单个N-连接的唾液酸化双触角聚糖。两个O-聚糖具有Gal β-1-3GalNAc 核心，其在末端Gal 处是α-2-3唾液酸化的。另外的二唾液酸化形式具有第二唾液酸残基，其与GalNAc 具有α-2-6键。单 -和二唾液酸化形式在人纤溶酶原中以 80：20的摩尔比发生。
纤溶酶原(PLG)基因定位于人类染色体6q26上。纤溶酶原蛋白物的分子量为?90kDa。它由在N端的Pan-苹果结构域以及C端的五个环状结构域和丝氨酸蛋白酶域组成。
Application
Sigma的酶已用于检查鼻腔分泌物中的纤溶酶原促进金黄色葡萄球菌表面菌丝化的能力。它已用于使用酶联免疫吸附法评估重组OspC蛋白与纤溶酶原的结合情况。其还可用于通过固相测定筛选 FITC标记的肺炎链球菌用于蛋白质相互作用。在这个测定中；用纤溶酶原包被孔，并与FITC标记的细菌一起孵育。在荧光计中读取荧光信号。

Biochem/physiol Actions
纤溶酶原是蛋白酶纤溶酶的无活性前体。纤溶酶原由组织型纤溶酶原激活剂 (tPA) 激活，tPA 主要激活纤溶酶的纤维蛋白溶解（血栓溶解）活性，或者由尿激酶型纤溶酶原激活剂 (uPA) 激活，uPA 与细胞外基质重塑和细胞迁移有关。纤溶酶可裂解纤维蛋白/纤维蛋白原和凝血因子 V/Va 以及 VIII/VIIIa。它可通过裂解非活性酶原激活基质金属蛋白酶。还与某些生长因子的激活有关，如血管內皮生长因子 (VEGF) 和转化生长因子 β (TGF-β)。
Preparation Note
溶液应储存在含有20mM 赖氨酸的中性 pH 缓冲液中。蛋白质应尽可能浓缩。在-20°C下等分冷冻，应可以稳定保存数周。
