一般描述

磷酸二酯酶(PDE)II属于PDE超家族（包括PDE1至PDE11亚家族）。以二聚体形式存在，由N端调节结构域、C端异戊二烯化结构域和一个含Zn2+的催化结构域组成。牛脾PDE2的分子量为65kDa。在二价阳离子的存在下发挥酶活性，最适pH为7.5。

应用

本品已用于多聚核苷酸链长、碱基组成和末端核苷酸的鉴定。酶已用于切除DNA中的吡啶氧丁基(POB)碱基加合物。另外还可联用微球菌核酸内切酶将纯化DNA水解为3-核苷单磷酸。
牛脾来源的磷酸二酯酶II已用于：
  • 切除DNA中的吡啶氧丁基(POB)碱基加合物
  • 在富集环腺苷之前消化DNA
  • 将蓝口贝鳃组织的DNA水解为脱氧核糖核苷3′-单磷酸

磷酸二酯酶（PDE）是一种可用于破坏磷酸二酯键的酶。 它是一种膜结合糖蛋白，可用于催化各种核苷酸多磷酸盐的水解。 磷酸二酯酶II已用于P8-dGMP复合物等纯化蛋白的酶法消化。 牛脾磷酸二酯酶已用于消化N-钙粘蛋白。

生化/生理作用

磷酸二酯酶(PDE)可破坏磷酸二酯键。3-异丁基-1-甲基黄嘌呤(IBMX)是一种潜在的PDE2抑制剂。
该酶作用于 poly (A)、poly (U) 和 poly (I)。天然 DNA 和 poly (C) 对这种酶的操作相当耐受。
将 RNA、RNA-Core、3'-烷基和 3'-芳基-核苷磷酸盐以及带有 3'-磷酸基团的多聚脱氧核糖核苷酸水解为 3'-单核苷酸。
不攻击具有 5'-磷酸单酯末端基团的多核苷酸。

单位定义

一个单位将产生相当于 aδ 的酸溶性核苷酸37°C、pH 6.5、2.0 mL 反应混合液中 A 260 /16 (30 分钟)底物：RNA-Core。用乙酸双氧铀-高氯酸试剂沉淀未水解 RNA 后，测定上清液中的实际 A 260 。
