一般描述
GenElute无内毒素质粒制备试剂盒可以简单、快速、低成本的纯化质粒DNA(内毒素水平≤0.1 EU/μg),所得产物可用于高效转染。 内毒素(也称为脂多糖或LPS)常常会与质粒DNA共纯化,这会显著降低内毒素敏感细胞系的转染效率。
取过夜培养物离心收集重组大肠杆菌,采用改良的SDS碱裂解法处理得到澄清的裂解液。通过简单的萃取相分离步骤去除内毒素。在高盐环境下,质粒DNA吸附到硅胶膜上接受进一步的纯化。简单的离心洗涤后,用无内毒素的水洗脱吸附的质粒DNA。25-200 mL培养物可以制备出多达1.2 mg不含内毒素的质粒DNA。
回收的质粒DNA主要以超螺旋形式存在。此类DNA可直接用于转染、限制性酶切、克隆、测序和PCR扩增等下游应用。
