General description

GenElute无内毒素质粒制备试剂盒可以简单、快速、低成本的纯化质粒DNA（内毒素水平≤0.1 EU/μg），所得产物可用于高效转染。 内毒素（也称为脂多糖或LPS）常常会与质粒DNA共纯化，这会显著降低内毒素敏感细胞系的转染效率。



取过夜培养物离心收集重组大肠杆菌，采用改良的SDS碱裂解法处理得到澄清的裂解液。通过简单的萃取相分离步骤去除内毒素。在高盐环境下，质粒DNA吸附到硅胶膜上接受进一步的纯化。简单的离心洗涤后，用无内毒素的水洗脱吸附的质粒DNA。25-200 mL培养物可以制备出多达1.2 mg不含内毒素的质粒DNA。

回收的质粒DNA主要以超螺旋形式存在。此类DNA可直接用于转染、限制性酶切、克隆、测序和PCR扩增等下游应用。
