General description
Synaptosomal-associated protein, 23 kDa, syndet (SNAP23), belonging to the SNAP25 family genes is a non-neuronal isoform of synaptosomal-associated protein 25 (SNAP-25), originally identified in a human B-lymphocyte cDNA library in a yeast two-hybrid screen for proteins interacting with syntaxin 4. SNAP-23 is ubiquitously expressed and is localized mainly to the plasma membrane. The primary structure of SNAP-23 is 59% identical to SNAP-25. It contains a central cluster of cysteine residues and predicted coiled-coil segments. It is a tSNARE which has two tandem soluble N-ethylmale-imide-sensitive factor-attachment protein receptor (SNARE) motifs. SNAP23 gene has eight exons and is located on human chromosome 15q21−22.
Immunogen
synthetic peptide corresponding to amino acids 2-20 of mouse SNAP-23, conjugated to KLH.This sequence is highly conserved in rat (90% identity) and human (78% identity) SNAP-23.
Application
Anti-SNAP-23 (DS-19) antibody produced in rabbit may be used in immunoblotting and immunofluorescence.
Biochem/physiol Actions
Anti-SNAP-23 (DS-19) recognizes SNAP-23 (23 kDa).
Synaptosomal-associated protein, 23 kDa, syndet (SNAP23) plays a vital role in various protein trafficking events in non-neuronal cells. It is mainly involved in polarized protein trafficking, glucose transporter type 4 (GLUT4) trafficking in adipocytes, compound exocytosis in mast cells and platelet dense core granule release. This gene acts as an oncogene in ovarian cancer (OC). This gene is capable of producing two forms of message, such as the full length message (SNAP23A) and a shorter message (SNAP23B).
Physical form
Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.
Preparation Note
For continuous use, store at 2-8°C for up to one month. For extended storage freeze in working aliquots. Repeated freezing and thawing is not recommended. Storage in "frost-free" freezers is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilution samples should be discarded if not used within 12 hours.
