General description
程序性细胞死亡1蛋白(PDCD1)定位于人类染色体2q37.3。PDCD1属于CD28受体家族，具有免疫抑制结构域。它在T和B淋巴细胞中表达。
细胞介导的免疫应答由T淋巴细胞启动，T淋巴细胞本身则由与抗原呈递细胞（APC）上的MHC分子结合的同源肽激活。T细胞的激活通常是自我限制的，因为激活的T细胞能够表达如PD-1（也被称为PDCD-1）等受体，其可介导来自APC的抑制信号。PD-1可以结合两种不同但相关的配体PDL-1和PDL-2。在与这些配体中的任一种结合后，PD-1产生的信号在没有“危险信号”（例如LPS或与细菌或其他病原体相关的其他分子）的情况下可抑制免疫应答的激活。有证据表明PD1缺失小鼠表现出过度活化的免疫系统和自身免疫疾病。与其预测的分子量不同，PD-1通常在SDS-PAGE中表现出更大的分子量迁移。
