General description

GTPase-activating protein and VPS9 domain-containing protein 1 (GAPVD1) also called Rab5-activating protein 6 (RAP6) or Gapex-5 gene is mapped to the human chromosome 9q33.3. It has a carboxy-terminal Vps9-like domain and an amino-terminal Ras GTPase-accelerating proteins (GAP)-like domain. GAPVD1 is present in the plasma membrane and endosomes.

Application

Anti-Gapex-5 antibody produced in rabbit may be used in immunoblotting and immunoprecipitation.

Biochem/physiol Actions

Anti-Gapex-5 recognizes human Gapex-5.
Rab5-activating protein 6 (RAP6) or Gapex-5 is an evolutionary conserved protein and its C-terminal Vps9 domain is responsible for the interaction with guanosine diphosphate GDP bound Rab5 and an N-terminal Ras GTPase-accelerating proteins (GAP) domain. Gapex-5 has guanine nucleotide exchange factor (GEF) activity specific for Rab5 and GAP activity specific for Ras. It is involved in multiple processes including receptor-mediated endocytosis, insulin receptor internalization and glucose transporter type 4 (Glut4) trafficking in adipocytes. RAP6 is also essential for epidermal growth factor receptor (EGFR) trafficking and degradation and phagosome maturation.

Physical form

Solution in 0.01 M phos­phate buffered saline, pH 7.4, containing 15 mM sodium azide.

Preparation Note

Store at –20 °C. For continuous use, the product may be stored at 2–8 °C for up to one month. For extended storage, freeze in working aliquots at –20 °C. Storage in “frost-free” freezers is also not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilutions should be discarded if not used within 12 hours.
