Dianhydrogalactitol is a blood brain barrier penetrant and potent alkylating agent that causes N7-guanine alkylation and inter-strand DNA crosslinks that induces cell cycle arrest followed by apoptosis. Dianhydrogalactitol exhibits potent antitumor activity in number of clinical models including metastatic NSCLC resistant to TKI and glioblastoma multiforme.
