AO-159 is a cell penetrant and potent pan-HMT (pan- Histone Methyltransferase) inhibitor that effectively inhibit the enzymatic activity of both G9a and Suv39H1 enzymes. AO-159 potently reduces the levels of H3K9me1, H3K9me2, and H3K9me3 in COS7 cells. In inhibits the five histone H3K9 methylating enzymes Suv39H1, Suv39H2, SETDB1, G9a, and GLP at low micromolar concentrations.
