Dynapyrazole Compound 20 is a potent and selective inhibitor of cytoplasmic S. cerevisiae dynein, an AAA (ATPase associated with diverse cellular activities), which engages the regulatory ATPase sites in the AAA3 and AAA4 domains disrupting allosteric communication across the AAA ring. Ddynapyrazole Compound 20 inhibits dynein’s basal ATPase activity. Also, it inhibits dynein motility by reducing the protein’s ability to bind microtubules.
