Org 34167 is a brain penetrant, potent and selective modulator of hyperpolarization-activated cyclic nucleotide-gated (HCN) channels that exhibit antidepressant activity in human and rat models. Apparently, Org 34167 inhibits HCN1 channel activity by modulating hyperpolarizing shift in activation voltage dependence and a slowing of activation kinetics.
