Biochem/physiol Actions
(R)-Ketamine (Arketamine), whose metabolite (2R,6R)-hydroxynorketamine exhibits AMPAR-modulating activity, contributes mainly to ketamine′s antidepressant efficacy. (S)-ketamine (Esketamine), on the other hand, is an NMDAR phencyclidine (PCP)-site antagonist that contributes to ketamine′s analgesic efficacy. Studies in rats and mice show that (R)-Ketamine displays the highest antidepressant potency and the least side-effects when compared with the racemate and (S)-ketamine.
