Cell penetrant, potent and selective first-in-class functional inhibitors of SLC15A4.
AJ2-30 is a cell penetrant, potent and selective first-in-class functional inhibitors of SLC15A4 (IC50 = 1.8 μM (IFN-α suppression) and IC50 = 2.6 μM (MDP transport inhibition)) that disrupt all functions of SLC15A4. AJ2-30 potently suppressed IFN-α production in human pDCs after stimulation with TLR7/8 and TLR9 agonists. It significantly suppressed production of IFN-γ, IL-6 and IL-10 in lupus patient-derived PBMCs, and significantly reduced IFN-α, IFN-β and IFN-γ production in mice.
