Fluorescent probe for dual autophagy and apoptosis induction detection in live cells.
CAQ2 is a dual autophagy and apoptosis probe with pH-dependent red fluorescent emission, displaying enhanced red emission upon lysosomal acidification during autophagy and drastically decreased red fluorescence in response to lysosomal alkalization during apoptosis. CAQ2 allows the visualization of autophagy (under starvation and lipidless conditions) and apoptosis induction (by UV irradiation, H2O2 and rotenone treatment) in live MCF-7 cells (cells are incubated with 5 μM CAQ2 for 30 min prior to red (Ex 488 nm, Em 530-630 nm) to blue (Ex 405 nm, Em 425-475 nm) fluorescence emission ratio detection/determination).
