Uniquely acting, highly selective sub-type activator for the KCNQ2 (KV7.2) potassium channel.
Ebio1 is a potent, sub-type selective KCNQ2 (KV7.2) potassium channel direct activator that is distinct from previously known activators of voltage-gated ion channels. Ebio1 operates through a unique mechanism, inducing a twist-to-open movement in the S6 helices of the channel, thereby increasing both the open probability and conductance of KCNQ2. KCNQ2 Activator Ebio1 has an EC50 of 247.3 nM for KCNQ2, making it tenfold more selective compared to KCNQ2/3, KCNQ4, and KCNQ5. Moreover, Ebio1 demonstrates minimal to negligible activation effects on other tested Kv, Nav, Cav, and K2P channel members, including hERG, BK, Nav1.1, Cav2.1, and TREK1.
