General description
L-ANAP (ANAP) is a bright fluorescent amino acid that can be site-specifically incorporated into proteins using amber stop codon suppression.
Application
L-ANAP may be used to fluorescently label proteins and study its localization and dynamics within live mammalian cells.
Biochem/physiol Actions
L-ANAP (ANAP), a fluorescent structural probe, enables real-time monitoring of ion channel dynamics through site-specific incorporation and environmental sensitivity. ANAP features optimal spectral properties with λex ~ 360 nm and environment-dependent λem ~ 490 nm, Φ of 0.22–0.5, and an ϵ of 17.5 mM−1 cm−1 in water. Due to its environmental sensitivity and ability to serve as a FRET donor, ANAP enables simultaneous monitoring of protein conformational changes, ligand binding, protein-protein interactions, and temperature sensitivity while maintaining channel function. ANAP’s versatility is demonstrated through successful applications in various cell types (Xenopus oocytes, HEK-293, CHO, tSA-201 cells), where it can be combined with electrical recordings to provide direct correlation between structural states and channel function. ANAP’s small size, environment-sensitive fluorescence, and the capability to introduce it at specific sites in the proteome through simple mutagenesis make it a unique and valuable tool for eukaryotic cell biology.
