20α-Hydroxycholesterol is a brain penetrant oxysterol present in human breast milk, human placenta and the mammalian CNS, which attenuates white matter injuries in neonatal mice. 20α-Hydroxycholesterol induces oligodendrogenesis through a sonic hedgehog Gli family zinc finger signaling activation mechanism. It increases numbers of mature oligodendrocytes, improves myelination, and rescues motor deficits.
