H-Gly-Phe-AFC trifluoroacetate salt

H-Gly-Phe-AFC (GF-AFC) is a cell-permeable dipeptidyl peptidase 1 (DPP1; cathepsin C; cathepsin J) substrate commonly used for cell viability accessment, where the fluorophore AFC is cleaved off by intracellular DPP1 activity in live cells. Unlike ATP and MTT assays, GF-AFC assay requires no lysis at the end of the assay, so cells can be used afterwards for other live cell-based assays. Cells are typically incubated with GF-AFC at a final concentration of 80 µM (from a 100 mM stock in DMSO) for 0.5-1 h at 37°C, and the AFC fluorescence intensity can be monitored by a fluorescence plate reader or by a fluorescence microscope (Ex 380-400 nm and Em 505 nm).
