STING degrader AK59 is a potent inhibitor of the cGAS/STING pathway (IC50 = 5.19 µM) by induction of STING interaction with ligase HERC4 (a HECT-domain E3 ligase) followed by degradation of STING. AK59 facilitates HERC4 to transfer ubiquitin directly to Lysine 150 of STING, which initiates the degradation. It is effective against wild-type STING and STING-associated vasculopathy (SAVI) mutant STING proteins in human CD14+ monocytic cells.
