Biochem/physiol Actions
Mito-HE is a mitochondria superoxide probe that corresponds to dihydroethidium covalently linked to the mitochondria-targeting triphenylphosphonium (TPP+) cation. Mito-HE is widely used for live cell imaging. Cells are incubated with the probe at a final concentration of 0.3-5 μM for 10-30 min at 37°C before washing. Two different excitation wavelengths are used to distinguish the superoxide-dependent hydroxylation of Mito-HE (385-405 nm) from the nonspecific formation of ethidium (480-520 nm). DMSO stock solutions should be stored at -20°C for no more than 3 weeks. Please refer to Nat Protoc. 2008 3(6):941-7 (PMID 18536642) for experimental setup recommendations.
