GCN2-IN-1 (A92; A-92; SRI-41964) is a potent GCN2 inhibitor (IC50 <300 nM; 20 min inhibitor preincubation prior to 90 min kinase reaction at RT with 10 μM ATP & 80 nM eIF2α) that effectively downregulates GCN2 phosphorylation in U2OS cells (GCN2 pSer51 IC50 <300 nM post 2h treatment). A92 is also reported to inhibit the RNA-binding protein HuR dimerization (IC50 = 4.5 μM).
