CAY10603, also known as BML-281 or Isox, is a highly selective HDAC6 inhibitor featuring an active site zinc-binding isoxazolo hydroxamate chelator, with IC50 values of 2.4 nM for HDAC6, 71 nM for HDAC1, and ~ 500 nM for HDAC10. CAY 10603 demonstrates significant anticancer potential, especially against pancreatic cancer cells with IC50 values below 0.5 μM, while also inhibiting tau deacetylation on KXGS motifs and Hedgehog signaling via Gli-dependent and independent pathways. CAY-10603 exhibits potent anti-inflammatory effects, suppressing inflammatory cascades and oxidative stress by modulating NF-κB and NLRP3 inflammasome pathways in various conditions.
