SC912 is a cell penetrant, potent and selective pan- androgen receptor (AR) inhibitor that binds to full-length AR as well as AR-V7 through AR N-terminal domain (IC50 = 0.36 and 1.01 μM, AR-V7 and ARv567es respectively). SC912 disturbs AR-V7 transcriptional activity and impairs AR-V7 nuclear localization and DNA binding. SC912 potently inhibits proliferation, induced cell-cycle arrest, and apoptosis of AR-V7 positive castration-resistant prostate cancer (CRPC) cells. It also inhibits growth of AR-V7 expressing CRPC xenografts.
