L-Homopropargylglycine hydrochloride

L-Homopropargylglycine (HPG) is a cell-permeable, non-canonical amino acid and structural analog of L-methionine, possessing a terminal alkyne (–C≡CH) group as a bioorthogonal handle. This structure is recognized by the native, wild-type methionyl-tRNA synthetase (MetRS), which mediates its incorporation into nascent polypeptide chains at methionine positions during ribosomal synthesis. Because MetRS promiscuously charges HPG onto tRNAᴹᵉᵗ in most cell types, this results in broad, non-specific labeling of the translatome. The incorporated alkyne handle serves as a versatile platform for covalent ligation via click chemistry, enabling either fluorescent visualization (FUNCAT) or affinity enrichment (BONCAT) of newly synthesized proteins. While lacking cell-type specificity, HPG provides an accessible and powerful tool for monitoring global proteome dynamics.
