L10, Copper Chelator

: L10 is a blood-brain barrier-permeable imine chelator that corrects copper dyshomeostasis in Alzheimer′s disease (AD) by targeting the pathogenic Cu-Aβ complex. L10 demonstrates functional efficacy by extracting nearly all Cu(II) from this complex in vitro at a 3:1 molar ratio. In a rat AD model, this mechanism translates to significant neuroprotection, marked by reduced neuroinflammation, oxidative stress, and Aβ accumulation in the hippocampal CA3. This targeted activity restores ATP7B transporter function and boosts BDNF expression, leading to a robust recovery of spatial memory. Distinguished by an excellent safety profile (EC50 = 914.3 µM) and optimized lipophilicity (LogP = 2.94), L10 is a compelling candidate for a disease-modifying AD therapy.
