AF64394 is a potent and selective inverse agonist for the G protein-coupled receptor GPR3 (IC50 = 0.5 µM GPR3, 7.9 µM GPR6 and 12 µM GPR12) that potently inhibits cAMP accumulation in HEK293 cells. It appears that AF64394 binds to the transmembrane dimer interface and functions as a negative allosteric modulator (NAM) specifically targeting dimeric GPR3.
