Mitoquinone mesylate (MitoQ), a TPP-CoQ10 conjugate, selectively accumulates 1,000-fold in the mitochondrial matrix via membrane potential (ΔΨm), redox cycling ubiquinol to ubiquinone, reducing ROS, upregulating endogenous antioxidants. Targeted delivery confers ~800-fold greater efficacy than untargeted antioxidants in preventing cell death, exhibiting antiproliferative effects in cancer (IC50s: 0.38 µM proliferation, 0.52 µM complex I inhibition). MitoQ provides systemic benefits (improved vascular/metabolic function, reduced liver inflammation/fibrosis), demonstrating chondrocyte cytotoxicity (1.094 mM) and efficacy in canine mammary gland tumor cells, offering a superior strategy combating organelle-specific oxidative stress and dysfunction across pathologies.
