Clonixeril (CXL) is a cell-permeable non-nucleotide that uniquely modulates hSTING, acting as either a weak partial agonist or a highly potent antagonist. Its antagonistic mechanism—blocking STING oligomerization and phosphorylation (Ser366)—is critically dependent on pre-agonist administration, as it cannot reverse pre-formed, disulfide-stabilized oligomers. Consequently, Clonixeril employs a non-classical, non-stoichiometric mechanism distinct from simple competitive antagonism.
