Application
Sphingosine kinase 2 is useful for the study of enzyme kinetics, screening inhibitors, and selectivity profiling. Sphingosine kinase 2 has been used in a study to assess the expression of sphingosine kinase 1 in oestrogen receptor-negative breast cancer. Sphingosine kinase 2 has also been used in a study to investigate the antitumor effects of sorafenib (≤ 10 μM) on four human neuroblastoma cell lines.
General description
Human Sphingosine kinase 2 (GenBank Accession No. AF245447), full length with N-terminal His tag, MW = 69.5 kDa, expressed in a Baculovirus infected Sf9 cell expression system.
Other Notes
One unit of sphingosine kinase activity is defined as the amount of enzyme required to produce 1 pmol of S1P/min
Physical form
Formulated in 25 mM Tris-HCl, pH 8.0, 100 mM NaCl, 0.05% Tween-20, 50% glycerol and 3 mM DTT.
Preparation Note
Thaw on ice. Upon first thaw, briefly spin tube containing enzyme to recover full content of the tube. Aliquot enzyme into single use aliquots. Store remaining undiluted enzyme in aliquots at -70°C. Note: Enzyme is very sensitive to freeze/thaw cycles.

Sphingolipids are a wide family of lipids that share common sphingoid backbones, including (2S,3R)-2-amino-4-octadecane-1,3-diol (dihydrosphingosine) and (2S,3R,4E)-2-amino-4-octadecene-1,3-diol (sphingosine). The metabolism and biological functions of sphingolipids derived from sphingosine have been the subject of many reviews. In contrast, dihydrosphingolipids have received

Neuroblastoma is the most common extracranial solid tumor in the pediatric population. Sorafenib (Nexavar), a multikinase inhibitor, blocks cell proliferation and induces apoptosis in certain types of cancers. Here, we tested antitumor effects of sorafenib (≤ 10 µM) on four

We previously reported that sphingosine 1-phosphate receptor 4 (S1P(4)) is expressed and stimulates the ERK-1/2 pathway via a human epidermal growth factor receptor 2 (HER2)-dependent mechanism in oestrogen receptor-negative (ER(-)) MDA-MB-453 breast cancer cells. Clinical relevance of S1P(4) and sphingosine
