General description
PLAU(纤溶酶原激活物,尿激酶)基因定位于人类染色体 10q22.2
尿激酶型纤溶酶原激活物又称 PLAU 和 uPA。人 uPA 最初合成为 431 个氨基酸前体,N 端信号肽(20 个残基)。单链分子被加工成不同分子量的二硫键连接的双链分子。从 Ser21(长型)和 Lys156(短型)开始,A 链以两种形式存在。长、短 A 链分别为高、低分子量形式所特有。长 A 链包含一个负责结合 PLAU 受体的EGF(表皮生长因子)样结构域。B 链对应于催化域。编码它的基因定位于10号人类染色体。
还原DTT。由于糖基化和裂解,蛋白质迁移成三条带,分别对应于长α链、β链和未处理的全长链。预测 N 端为Ser21, Ile179& Lys156。
Biochem/physiol Actions
尿激酶型纤溶酶原激活物是一种底物特异性极其有限的丝氨酸蛋白酶,其裂解纤溶酶原中的 Cys-Pro-Gly-Arg560-Val561-Val-Gly-Gly-Cys 序列从而形成纤溶酶。在与乳腺癌、胃癌、结肠癌、膀胱癌、卵巢癌、脑癌和子宫内膜癌相关的多种人类癌症中 ,uPA(纤维蛋白溶酶原激活物,尿激酶)介导的迁移细胞细胞外基质降解对组织重塑、浸润和血管生成至关重要。uPA 促进银屑病和基底细胞癌的发病。 uPA 激活生长因子和金属蛋白酶。
Physical form
用0.22μm 过滤的 HEPES、NaCl 和 CaCl2 溶液冻干。冻干前一般加入 5-8% 甘露醇或海藻糖作为保护剂。
Preparation Note
打开前离心小瓶。用无菌 PBS(pH 值为 7.4)重建至 50μg/ mL 的浓度。避免涡旋。该溶液可在 2-8°C 下储存长达 1 个月。对于长期存储,建议储存温度为 -20°C。
