General description
Clusterin is expressed in all tissues. However, it is strongly expressed in the central nervous system and plasma. Clusterin mainly protects against heat and other stress. It is also referred as chaperone protein and stress-inducible biomarker. Clusterin exists in two isoforms, one is a secretory form and other is a nuclear form. The secreted form of the protein is a 80kDa disulfide-linked heterodimer of α and β subunits (produced by internal cleavage). The alternatively spliced inactive cytosolic form is changed to a truncated mature nuclear isoform via ionizing irradiation.
Biochem/physiol Actions
Clusterin is involved in tissue remodeling, cell death, lipid transport, complement-associated cell lysis, cancer and neurological damage. It is strongly expressed in Alzheimer′s and treatment-resistant cancer. High levels of clusterin are observed in ischemic heart disease and diabetic conditions. It also protects kidney from fibrosis. Nuclear form of clusterin works as a pro-death signal, and suppresses cell growth and survival. The secreted form of clusterin has extracellular chaperone and anti-apoptotic activities.

Solution in 55 mM TRIS-HCl, pH 8.2, containing 150 mM NaCl.

Mouse nuclear clusterin (aa 22-448) is fused at the C-terminus to a His-tag.
