General description
Urocortin (UCN) is a neuropeptide, widely distributed in the central nervous system (CNS). Urocortin is derived from a larger precursor protein (pro-urocortin, also named corticotensin).
Urocortin (UCN) is encoded by the gene mapped to human chromosome 2p21–23. The encoded protein belongs to the corticotropin-releasing hormone (CRH) family. UCN is a 40 amino acid long peptide. It is widely expressed in the heart, lung, skin and immune cells.
Immunogen
synthetic peptide corresponding to the C-terminal of human urocortin (amino acids 25-40 with N-terminally added lysine) conjugated to KLH. The immunogen sequence (human) corresponds to pro-urocortin (amino acids 105-120) and is identical in rat urocortin.
Application
Anti-Urocortin antibody produced in rabbit has been used in:
- radioimmunoassay (RIA)
- immunohistochemistry
- immunocytochemistry
Anti-urocortin antibody produced in rabbit has been used to evaluate the entry of urocortin peptide into the brain.
Biochem/physiol Actions
The antibody is evaluated for activity and specificity by radioimmunoassay (RIA) and immunohistochemistry. The antibody does not cross-react with human or rat CRF (6-33), sheep CRF, PACAP38, human ACTH, human ACTH (18-39), and sauvagine.
Urocortin (UCN) plays a vital role in inhibiting transforming growth factor β1 (TGFβ1) oncogenic signaling pathway. The encoded protein is implicated in systemic and peripheral stress-response systems. Mutations in UCN gene might increase the risk of susceptibility to obesity.
Urocortin has biological actions like corticotropin-releasing factor (CRF) such as autonomic and behavioral responses in mammalian species. It is a potent stimulator of adrenocorticotropic hormone (ACTH) secretion by corticotroph cells from the anterior pituitary in vitro and in vivo. In the central nervous system (CNS), urocortin acts to produce potent anxiogenic-like and appetite suppressing effects and peripherally has hypotensive effects.
