一般描述

Extract-N-Amp组织PCR试剂盒为从各种细胞和组织中快速提取DNA并通过直接PCR(聚合酶链反应)扩增靶标提供了所有必需试剂。这种新型提取方法无需长时间的酶消化或均质化处理。该试剂盒还包括专门配制的热启动PCR ReadyMix,可直接从提取物中扩增。

组分

试剂盒含有Tissue Preparation Solution、Extraction Solution和Extract-N-Amp PCR Reaction Mix。

原理

该试剂盒包含从小鼠尾、毛发、动物组织、唾液和口腔拭子中提取和扩增基因组DNA的经验证实验方案。在常规操作步骤中,基因组DNA提取自在室温下的组织制备溶液和提取溶液中孵育10分钟的样本。将该样本加热至95 °C并持续3分钟,然后混入第三种溶液以便在PCR反应前中和抑制物。随后将一部分DNA提取物加入到含有引物和Extract-N-Amp PCR ReadyMix(包含在试剂盒内)的PCR反应物中。
