General description
The ZytoLight® SPEC MDM2/CEN 12 Dual Color Probe (PL18) is a direct‑labeled dual‑color fluorescence in situ hybridization (FISH) probe for detecting the human MDM2 gene region together with alpha‑satellite sequences of chromosome 12 in formalin‑fixed, paraffin‑embedded specimens.It combines differently labeled probe fractions targeting the MDM2 locus and chromosome 12 centromeric sequences to support visualization of copy number changes at these regions by FISH on tissue sections.
Principle:
The ZytoLight® SPEC MDM2/CEN 12 Dual Color Probe utilizes fluorescence in situ hybridization (FISH) to detect specific nucleic acid sequences in cell preparations from formalin-fixed, paraffin-embedded (FFPE) tissue. Fluorescently labeled DNA probes—ZyGreen for the MDM2 gene region (12q15) and ZyOrange for chromosome 12 alpha satellite (CEN 12, 12p11.1-q11)—co-denature with complementary target DNA, hybridize overnight, and have unbound fragments removed via stringency washing. Counterstaining with DAPI enables visualization of hybridized green (MDM2) and orange (CEN 12) signals using a fluorescence microscope with appropriate excitation/emission filters.

- ZyOrange: Labeled polynucleotides (excitation 547 nm/emission 572 nm) targeting sequences in 12p11.1-q11 (specific for the alpha satellite centromeric region D12Z3 of chromosome 12).
- Hybridization Buffer: Formamide-based hybridization buffer.
Application
The probe is designed for qualitative detection of translocations between 14q32.33 and 8q24.21 regions in formalin-fixed, paraffin-embedded (FFPE) tissue specimens by FISH. It is used in combination with the ZytoLight® FISH-Tissue Implementation Kit for specimen pretreatment, hybridization, and post-hybridization processing.

Analysis Note
The performance of the probe has been validated for detecting amplifications of the MDM2 gene locus. In normal interphase cells, two green signals (representing the MDM2 gene region) and two orange signals (representing the centromeric region of chromosome 12) are expected. In cases of amplification, multiple copies of the green signal or clusters will be observed.
