General description
The ZytoLight® SPEC CCND1 Dual ColorBreak Apart Probe (PL65) is a probe for qualitative detection of translocations associated the human CCND1 gene at 11q13.3.
Application
It specifically assists in differentiating atypical lipomatous tumors/well-differentiated liposarcomas (ALT/WDLPS) and dedifferentiated liposarcoma (DDLPS) by identifying CDK4 amplification, a crucial marker that distinguishes these tumors from other sarcomas.
Analysis Note
- With the use of appropriate filter sets, the hybridization signals of the probeappear green (proximal to the CCND1 breakpoint region) and orange(distal to the CCND1 breakpoint region).
- Normal situation: In interphases of normal cells or cells without atranslocation involving the CCND1 gene region, two green/orange fusionsignals appear
- Aberrant situation: One CCND1 gene region affected by a translocation isindicated by one separate green signal and one separate orange signal
- Overlapping signals may appear as yellow signals.
