General description
The ZytoLight SPEC ETV6 Dual Color Break Apart Probe (PL135) is a direct-labeled FISH probe designed for the qualitative detection of translocations involving the human ETV6 gene at 12p13.2 in formalin-fixed, paraffin-embedded (FFPE) specimens. It is intended for use with the ZytoLight FISH-Cytology Implementation Kit.
Components:
ZytoLight® SPEC ETV6 Dual Color Break Apart Probe comprises:
FISH employs direct-labeled probes flanking ETV6 breakpoint, co-denatured (72°C/5 min) with target DNA on pretreated cytology slides (10 µl/slide), hybridized overnight (37°C), stringency washed, DAPI counterstained, and visualized by fluorescence microscopy using ZytoLight FISH-Cytology Implementation Kit (Z-2099-20).
Components:
ZytoLight® SPEC ETV6 Dual Color Break Apart Probe comprises:
- ZyGreen labeled polynucleotides (excitation 503 nm/emission 528 nm) targeting sequences proximal to the ETV6 breakpoint region on chromosome 12 (12p13.2).
- ZyOrange labeled polynucleotides (excitation 547 nm/emission 572 nm) targeting sequences distal to the ETV6 breakpoint region.
- Formamide-based hybridization buffer.
FISH employs direct-labeled probes flanking ETV6 breakpoint, co-denatured (72°C/5 min) with target DNA on pretreated cytology slides (10 µl/slide), hybridized overnight (37°C), stringency washed, DAPI counterstained, and visualized by fluorescence microscopy using ZytoLight FISH-Cytology Implementation Kit (Z-2099-20).
Application
ZytoLight® SPEC ETV6 Dual Color Break Apart Probe aids in the differential analysis of conditions associated with ETV6 rearrangements, including ESS and other related contexts. It is used for the qualitative detection of translocations involving the ETV6 gene at 12p13.2 by fluorescence in situ hybridization (FISH)
Analysis Note
In a normal interphase nucleus, two green and two orange signals are expected. In cases of ETV6 rearrangements, an aberrant signal pattern may present as one orange/green fusion signal, one orange signal, and one separate green signal, indicating the presence of a translocation. Any unexpected signal patterns should be further investigated.
