Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Preparation Note

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
Reconstitute lyophilized antibody pellet with 25μL of ultrapure water or Phosphate Buffered Saline (PBS). Please refer to our reconstitution protocol and the specific application guidance on the suggested starting dilutions and sample type.

General description

Ras-related protein Rab-10 (UniProt: P61026; also known as Rab10 ) is encoded by the RAB10 gene (Gene ID: 10890) in human. Rab10, a member of the Rab family of small GTPases, is a key regulator of intracellular membrane trafficking, from the formation of transport vesicles to their fusion with membranes. It is mainly involved in the biosynthetic transport of proteins from the Golgi to the plasma membrane. It also plays a specific role in asymmetric protein transport to the plasma membrane. Rab10 localizes to the endoplasmic reticulum (ER) and to dynamic ER-associated structures that track along microtubules and mark the position of new ER tubule growth. Depletion of Rab10 alters ER morphology that results in fewer ER tubules. In the brain, Rab10 is expressed in all cell types. It is activated by phosphorylation at threonine 73 by LRRK2. The phosphorylated Rab10 is reported to colocalize with neurofibrillary tangles in Alzheimer s disease brains. In a cellular model of amyloid precursor protein (APP) metabolism, silencing of RAB10 gene is reported to lead to a significant reduction in Aβ42 and the Aβ42/40 ratio. Application

Quality Control Testing

Evaluated by Western Blotting in MCF-7 cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected Rab10 in MCF-7 cell lysate.

Tested applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected Rab10 in lysates fro A549 cells, Mouse brain tissue, and C6 cells.

Affinity Binding Assay: A representative lot of this antibody bound Rab 10 peptide with a KD of 1.4 x 10-8 in an affinity binding assay.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected Rab10 in human brain and mouse brain tissue sections.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user
