General description
Chromodomain-helicase-DNA-binding protein 8 (UniProt: Q9HCK8; also known as EC: 3.6.4.12, CHD-8, ATP-dependent helicase CHD8, Helicase with SNF2 domain 1) is encoded by the CHD8 (also known as HELSNF1, KIAA1564) gene (Gene ID: 57680) in human. CHD-8 is a member of the SNF2/RAD54 helicase family. Two isoforms of CHD-8 have been reported that are produced by alternative splicing. It is a DNA helicase that acts as a chromatin remodeling factor and regulates transcription. It also acts as a transcription repressor by remodeling chromatin structure and recruiting histone H1 to target genes. CHD-8 is reported to suppress p53/TP53-mediated apoptosis by recruiting histone H1 and preventing p53/TP53 transactivation activity and also acts as a negative regulator of Wnt signaling pathway by regulating beta-catenin activity. It negatively regulates β-catenin-targeted gene expression by being recruited specifically to the promoter regions of several β-catenin responsive genes. CHD-8 is involved in both enhancer blocking and epigenetic remodeling at chromatin boundary via its interaction with CTCF. Mutations in CHD8 gene are associated with Autism 18 that is characterized by impairments in reciprocal social interaction and communication and developmental abnormalities. Clone 1H9 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects Chromodomain-helicase-DNA-binding protein 8 (CHD-8). It targets an epitope within 20 amino acids from the internal region.
Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.
Preparation Note
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
Reconstitute lyophilized antibody pellet with 25μL of ultrapure water or Phosphate Buffered Saline (PBS). Please refer to our reconstitution protocol and the specific application guidance on the suggested starting dilutions and sample type.
