General description
Ribonuclease 3 (UniProt: Q9NRR4; also known as EC:3.1.26.3, Protein Drosha, Ribonuclease III, RNase III, p241) is encoded by the Drosha (also known as RN3, RNASE3L, RNASEN) gene (Gene ID: 29102) in human. Drosha is a ubiquitously expressed Ribonuclease III double-stranded (ds) RNA-specific endoribonuclease that is involved in the initial step of microRNA (miRNA) biogenesis. It is a component of the microprocessor complex, or pri-miRNA processing protein complex, which is composed of Drosha and DGCR8 (Pasha). Within this complex, Drosha cleaves the 3′ and 5′ strands of a stem-loop in pri-miRNAs to release hairpin-shaped pre-miRNAs that are subsequently cut by the cytoplasmic DICER to generate mature miRNAs. Drosha is shown to cleaves only the double-strand RNA and does not cleave single-strand RNA. The ectopic expression of Drosha is reported to play a significant role in cancer progression and is observed in a variety of tumors. Higher expression of Drosha in reported in non-small cell lung cancer, squamous cell carcinomas, and basal cell carcinomas. It is associated with advanced stage of these tumors. Overexpression of Drosha is observed in the adrenocortical carcinomas. Clone 1B19 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects DROSHA. it targets an epitope with 22 amino acids from the N-terminal region.
Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.
Preparation Note
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
Reconstitute lyophilized antibody pellet with 25μL of ultrapure water or Phosphate Buffered Saline (PBS). Please refer to our reconstitution protocol and the specific application guidance on the suggested starting dilutions and sample type.
