General description

ZooMAb® antibodies represent an entirely new generation of recombinant monoclonal antibodies. Each ZooMAb® antibody is manufactured using our proprietary recombinant expression system, purified to homogeneity, and precisely dispensed to produce robust and highly reproducible lot-to-lot consistency. Only top-performing clones are released for use by researchers. Each antibody is validated for high specificity and affinity across multiple applications, including its most commonly used application. ZooMAb® antibodies are reliably available and ready to ship when you need them.

G1/S-specific cyclin-E2 (UniProt: O96020) is encoded by the CCNE2 gene (Gene ID: 9134) in human. The G1/S-specific cyclin-E2 (cyclin-E2) is a critical regulator of the cell cycle and has garnered significant attention due to its roles in cell proliferation, DNA replication, and its implications in oncogenesis. Cyclin-E2 is highly homologous to cyclin-E1, with approximately 50% sequence identity, and contains conserved domains such as the cyclin box, nuclear localization signals (NLS), and centrosome localization signals (CLS). Cyclin-E2 interacts with cyclin-dependent kinase 2 (CDK2), forming a kinase complex that drives the G1/S phase transition by phosphorylating substrates like retinoblastoma protein (Rb), E2F transcription factors, and other components of the pre-replication complex (PRC). Cyclin-E2 expression peaks during the G1/S transition, regulated predominantly by E2F transcription factors, which are released from Rb upon phosphorylation by CDK4/6-cyclin-D complexes. Cyclin-E2/CDK2 activity modulates critical cellular processes, including initiation of DNA replication, centrosome duplication, and histone biosynthesis. Unlike cyclin-E1, cyclin-E2 is implicated in distinct regulatory functions, including interactions with centrosomic proteins and potential roles in cellular stress response pathways. Cyclin-E2 localizes to the nucleus during late G1 and early S-phase, aligning with its role in transcriptional regulation and DNA replication initiation. Cyclin-E2 degradation is mediated by the SCF-FBW7 ubiquitin ligase complex, ensuring tight regulation throughout the cell cycle. Dysregulation of cyclin-E2, often through gene amplification or FBXW7 mutations, is frequently observed in cancers, contributing to replication stress, genomic instability, and resistance to therapies. Elevated cyclin-E2/CDK2 activity correlates with poor prognosis across multiple tumor types. This ZooMAb® recombinant monoclonal antibody, generated by our proprietary technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.

Immunogen

KLH-conjugated linear peptide corresponding to 23 amino acids from the N-terminal half of human Cyclin E2.

Application

Quality Control Testing

Evaluated by Western Blotting in Jurkat cell lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected Cyclin E2 in Jurkat cell lysate.

Tested Applications

Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected Cyclin E2 in L6 cell lysate.

Affinity Binding Assay: A representative lot of this antibody bound with Cyclin E2 peptide with at least ten thousand-fold (10,000X) higher affinity than with non-specific control peptide.

Immunoprecipitation Analysis: 2 µg from a representative lot immunoprecipitated Cyclin E2 from Jurkat cell lysate.

Note: Actual optimal working dilutions must be determined by end user as specimens and experimental conditions may vary with the end user.

Biochem/physiol Actions

Clone 1F10 is a ZooMAb® rabbit recombinant monoclonal antibody that detects Cyclin E2. It targets an epitope within 23 amino acids from the N-terminal half.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Preparation Note

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
