NF-kappa-B essential modulator (UniProt: Q9Y6K9; also known as NEMO, NF-kappa-B essential modulator, FIP-3, IkB kinase-associated protein 1 (IKKAP1), Inhibitor of nuclear factor kappa-B kinase subunit gamma (I-kappa-B kinase subunit gamma; IKK-gamma; IKKG; IkB kinase subunit gamma), NF-kappa-B essential modifier) is encoded by the IKBKG (also known as FIP3, NEMO) gene (Gene ID: 8517) in human. NF-kappa-B essential modulator (IKK ) is a crucial component of the I-kappa-B kinase (IKK) complex, which plays a pivotal role in the NF-kappa-B signaling pathway. IKBKG is part of the IKK core complex, consisting of CHUK, IKBKB, and IKBKG, forming a structure that facilitates the phosphorylation and subsequent degradation of I-kappa-B proteins, thereby allowing NF-kappa-B to translocate to the nucleus and activate gene transcription. IKK is ubiquitously expressed across various tissues, highlighting its fundamental role in immune response regulation. Mutations or dysregulation of IKK are associated with several diseases, including Incontinentia Pigmenti, a genetic disorder characterized by skin, dental, and neurological abnormalities, and various immunodeficiencies due to impaired NF-kappa-B activation. Recent studies have also implicated IKK in cancer progression and inflammatory diseases. Clone 1L20 is a ZooMAb® rabbit recombinant monoclonal antibody that detects IKK . It targets an epitope within 15 amino acids from the internal region.

应用

Quality Control Testing

Evaluated by Western Blotting in A-172 cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected IKK in A-172 cell lysate.

Tested Applications

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected IKK in human placenta and human pancreas tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound with IKK peptide with at least ten thousand-fold (10,000X) higher affinity than with non-specific control peptide.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

外形

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
