Signal transducer and activator of transcription 5B (UniProt: P51692; also known as STAT5B) is encoded by the STATB gene (Gene ID: 6777) in human. STAT (Signal Transducers and Activators of Transcription) proteins are a family of conserved transcription factors that transduce high-fidelity signals for the cytokine family of ligands and serve as principal substrates of Janus kinases (JAK). In resting cells, STATs exist in an inactive state in the cytoplasm until phosphorylated and activated. Activators include cytokines, growth factors, and some peptides. STAT5B carries out the dual function of signal transduction and activation of transcription. It mediates cellular responses to the KIT ligand/SCF and other growth factors. Binds to the -interferon activation site (GAS) element and activates PRL-induced transcription. It is known to positively regulate hematopoietic/erythroid cell differentiation. Following tyrosine phosphorylation in response to KIT signaling, it is activated and translocates to the nucleus. It can also be phosphorylated by JAK2. Its phosphorylation at tyrosine 699 by protein tyrosine kinase 6 (PTK6) or Hematopoietic cell kinase (HCK) leads to an increase in its transcriptional activity. Mutations in STAT gene have been linked to large granular lymphocytic (LGL) leukemia. The Y665F and N642H mutants show increased transcriptional activity and tyrosine 694 phosphorylation. Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.

Reconstitute lyophilized antibody pellet with 25μL of ultrapure water or Phosphate Buffered Saline (PBS). Please refer to our reconstitution protocol and the specific application guidance on the suggested starting dilutions and sample type.

Quality Control Testing

Evaluated by Western Blotting in MOLT4 cell lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected STAT5B in MOLT4 cell lysate.

Tested Applications

Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected STAT5B in lysate from Daudi cells and 1:1,000 dilution in lysate from HeLa cells.

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected STAT5B in human placenta and human tonsil tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound STAT5B peptide with a KD of 1.0 x 10-6 in an affinity binding assay.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected STAT5B in Daudi cells.
