General description
Mitotic checkpoint serine/threonine-protein kinase BUB1 beta (UniProt: O60566), also known as Mitotic MAD3/BUB1-related protein kinase or budding uninhibited by benzimidazole 1-related 1 (BUBR1). It is encoded by the BUB1B gene (Gene ID: 701) in human. BUBR1 is a serine/threonine protein kinase and a member of the BUB1 family. It consists of multiple functional domains, including a kinase domain responsible for catalytic activity as well as domains involved in binding to other mitotic checkpoint proteins and kinetochores. BUBR1 is an essential component of mitotic checkpoint complex (MCC). With other proteins it coordinates and monitor the attachment of chromosomes to the mitotic spindle and prevents premature cell division until all chromosomes are correctly aligned at the metaphase plate. BUBR1 also involved in regulating the activity of the anaphase-promoting complex/cyclosome (APC/C), a key regulator of cell cycle progression and degradation of specific proteins. Furthermore, BUBR1 contributes to DNA damage response pathways and mitotic exit control. BUBR1 expression is tightly regulated throughout the cell cycle, with peak levels during metaphase and decreased expression during anaphase and telophase in actively dividing cells during mitosis. It is particularly abundant in tissues with high cell turnover rates such as testes, thymus, and bone marrow. BUBR1 gene mutation or dysregulation can result in chromosomal instability, aneuploidy, and defective chromosome segregation, which are hallmarks of cancer development and progression. BUBR1 is downregulated with age and associated with age-related disorders. Clone 1D23 is a ZooMAb® rabbit recombinant monoclonal antibody that detects Mitotic checkpoint serine/threonine-protein kinase BUB1β (BubR1). It targets an epitope within 25 amino acids from the internal region.
Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
Preparation Note
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
Reconstitute lyophilized antibody pellet with 25μL of ultrapure water or Phosphate Buffered Saline (PBS). Please refer to our reconstitution protocol and the specific application guidance on the suggested starting dilutions and sample type.
