Western Blotting Analysis: A 1:10,000 dilution of this antibody detected CDLK5 in in HeLa cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected CDKL5 in lysate from Mouse brain tissue and 1:10,000 in Rat brain tissue.

Affinity Binding Assay: A representative lot of this antibody bound CDKL5 peptide with a KD of 5.6 x 10-9 in an affinity binding assay.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected CDKL5 in Human colon tissue sections.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected CDKL5 in HeLa cells.

Clone 2J15 is a ZooMAb® rabbit recombinant monoclonal antibody that detects Cyclin-dependent kinase-like 5 (CDKL5). It targets an epitope within 23 amino acids in the C-terminal half.

General description

Cyclin-dependent kinase-like 5 (UniProt: O76039) is encoded by the CDKL5 gene also known as STK9 (Gene ID: 6792) in human. It is a Serine/Threonine-protein kinase that mediates phosphorylation of MECP2. The Primary structure of Cyclin-dependent kinase-like 5 protein consists of an N-terminal catalytic domain, including the TEY sequence observed in MAPK family kinases, a C-terminal regulatory domain, two nuclear localization signal (NLS) sequences and a nuclear export signal (NES). Subcellular localization of cyclin-dependent kinase-like 5 is regulated through phosphorylation by DYRK1A on Ser-308 residue near the NLS (amino acids 312 315). As a result, the protein is exported to the cytoplasm. High expression of Cyclin-dependent kinase-like 5 protein is observed either in the nucleus or in the cytoplasm in various brain areas and developmental stages. It plays a role in brain development, neuronal maturation and migration, and synapse formation among others. Mutations or chromosomal translocations in the CDKL5 gene are associated with neurodevelopmental disorders like Cyclin-dependent kinase-like 5 (CDKL5) deficiency disorder (CDD) and developmental and epileptic encephalopathy 2, and found in patients manifesting a severe form of epilepsy, early-onset seizures, mental retardation and in West syndrome. Immunogen

KLH-conjugated linear peptide corresponding to 23 amino acids within the C-terminal half of Human Cyclin-dependent kinase-like 5 (CDKL5).

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Preparation Note

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
Reconstitute lyophilized antibody pellet with 25μL of ultrapure water or Phosphate Buffered Saline (PBS). Please refer to our reconstitution protocol and the specific application guidance on the suggested starting dilutions and sample type.
