Western Blotting Analysis: A 1:1,000 dilution of this antibody detected SPARC in C6 cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected His-tagged, recombinant fragment of human SPARC.

Flow Cytometry Analysis: 1 µg from a representative lot detected SPARC in one million Human umbilical vein endothelial cells (HUVEC).

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected SPARC in Human umbilical vein endothelial cells (HUVEC).

Immunohistochemistry (Paraffin) Analysis: A 1:1000 dilution from a representative lot detected SPARC in human kidney tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound SPARC peptide with at least ten million-fold (10,000,000X) higher affinity than with non-specific control peptide.

Clone 5N19 is a ZooMAb^® recombinant rabbit monoclonal antibody that specifically detects SPARC. It targets an epitope within 24 amino acids from the C-terminal half.

Basement-membrane protein 40 (BM-40) (UniProt: P09486; also known as Osteonectin, ON, Secreted protein acidic and rich in cysteine) is encoded by the SPARC (also known as ON) gene (Gene ID: 6678) in human. It is a matrix-associated protein that elicits changes in cell shape, inhibits cell-cycle progression, and influences the synthesis of extracellular matrix (ECM). It also binds to calcium, copper, several types of collagen, albumin, thrombospondin, PDGF and cell membranes. SPARC protein has an acidic domain that binds 5 to 8 Ca2+ with a low affinity, a cysteine-rich domain, and an EF-hand loop that binds a Ca2+ ion with a high affinity. The protein is synthesized with a signal peptide (aa 1-17), which is subsequently cleaved off to produce the mature form (aa 18-303). It is expressed at high levels in tissues undergoing morphogenesis, remodeling and wound repair. SPARC protein is primarily secreted into the extracellular space, where it interacts with the extracellular matrix. Although, being highly valued in non-nerve tissues, studies in both cell and animal models have revealed that SPARC family proteins may also continue to play a vital role in central nervous system (CNS) diseases and development. SPARC has also been demonstrated as an important player in the context of obesity, diabetes, and fatty liver disease including advanced hepatic fibrosis and hepatocellular carcinoma. Evidence in murine and human samples indicates that SPARC is involved in adipogenesis, cellular metabolism, extracellular matrix modulation, glucose and lipid metabolism. Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
