ZooMAb® antibodies represent an entirely new generation of recombinant monoclonal antibodies. Leptin (UniProt: P41159; also known as Obese protein, Obesity factor) is encoded by the LEP (also known as OB, OBS) gene (Gene ID: 3952) in human. Leptin is an adipocyte-derived hormone pivotal in regulating energy homeostasis, appetite, and metabolism. Leptin belongs to the cytokine superfamily and is composed of a helical structure resembling IL-6. It binds to the leptin receptor (LEP-R), a member of the class I cytokine receptor family, initiating downstream signaling pathways such as JAK-STAT, PI3K, and MAPK. The receptor-ligand interaction occurs predominantly in the hypothalamus, where leptin suppresses appetite and regulates energy expenditure. Leptin acts as a key regulator of body weight by signaling satiety and reducing food intake. It is secreted by white adipose tissue and circulates in the bloodstream, with its levels positively correlating with fat mass. Beyond energy regulation, leptin influences immune function, hematopoiesis, and reproduction. Dysregulation in leptin signaling, often manifesting as leptin resistance, is implicated in obesity, diabetes, and metabolic syndrome. Additionally, leptin modulates cytokine production and inflammatory responses, linking its role to infectious diseases and autoimmune conditions. Leptin receptor expression is widespread, including in endocrine tissues like the ovary, uterus, and testis, as well as in the hypothalamus and cardiovascular system. Aberrant leptin signaling pathways have been associated with cardiac dysfunction and impaired cellular processes, highlighting its importance in maintaining systemic homeostasis. Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected the full length, recombinant human Leptin protein and a 1:1,000 dilution detected Leptin in HaCaT cell lysate.
Affinity Binding Assay: A representative lot of this antibody bound with Leptin peptide with at least ten thousand-fold (10,000X) higher affinity than with non-specific control peptide.
Enzyme Immunoassay: A variety dilutions of representative lot of this antibody detected Leptin protein in ELISA application.
Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Clone 1D23 is a ZooMAb® rabbit recombinant monoclonal antibody that detects Leptin. It targets an epitope within 21 amino acids from the internal region.
外形
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
