General description

ZooMAb® antibodies represent an entirely new generation of recombinant monoclonal antibodies. Each ZooMAb® antibody is manufactured using our proprietary recombinant expression system, purified to homogeneity, and precisely dispensed to produce robust and highly reproducible lot-to-lot consistency. Volume-regulated anion channel subunit LRRC8B (UniProt: Q6P9F7; also known as Leucine-rich repeat-containing protein 8B, T-cell activation leucine repeat-rich protein (TA-LRRP)) is encoded by the LRRC8B (also known as KIAA0231, UNQ6413/PRO21207) gene (Gene ID: 23507) in human. LRRC8B is a subunit of the volume-regulated anion channel (VRAC), a heteromeric protein complex essential for maintaining cellular volume homeostasis. It features transmembrane domains and leucine-rich repeats, which contribute to its role in ion transport. It forms functional VRACs through heteromerization with LRRC8A, the essential subunit, and other LRRC8 paralogs (LRRC8C-E). These channels are activated by hypoosmotic cell swelling and facilitate the transport of chloride ions and organic osmolytes, such as taurine and glutamate, across the plasma membrane. LRRC8B is expressed in various tissues and is primarily localized to the plasma membrane, although it also appears in the endoplasmic reticulum (ER) under certain conditions. The subunit composition of VRACs, including LRRC8B, governs their ion selectivity, conductance, and regulatory properties. Beyond its role in osmotic regulation, LRRC8B has been implicated in calcium homeostasis and intracellular signaling pathways. For instance, its absence has been shown to impair calcium release from the ER, thereby influencing cellular calcium dynamics. Dysfunction or altered expression of LRRC8B has been associated with diseases such as megalencephalic leukoencephalopathy with subcortical cysts and spinocerebellar ataxia 44. Recent studies also highlight its potential roles in cancer biology, where it may influence tumor cell survival and proliferation through its effects on ionic and osmotic balance. Immunogen

KLH-conjugated linear peptide corresponding to 25 amino acids from the N-terminal cytoplasmic domain of human LRRC8B.

Application

Quality Control Testing
Evaluated by Western Blotting in wild-type HeLa cell lysate. 
Western Blotting Analysis: A 1:1,000 dilution of this antibody detected LRRC8B in wild-type HeLa cell lysate, but not in LRRC8B knockout HeLa cell lysate. 
Tested Applications
Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected LRRC8B in human brain tissue lysate and a 1:10,000 dilution from HEK-293 and human umbilical vein endothelial cell (HUVEC) lysates.
Immunoprecipitation Analysis: 2 µg from a representative lot immunoprecipitated LRRC8B from human umbilical vein endothelial cell (HUVEC) lysate.
Affinity Binding Assay: A representative lot of this antibody bound with LRRC8B peptide with at least one hundred thousand-fold (100,000X) higher affinity than with non-specific control peptide.
Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Biochem/physiol Actions

Clone 1C14 is a ZooMAb® rabbit recombinant monoclonal antibody that detects LRRC8B. It targets an epitope within 25 amino acids from the N-terminal, cytoplasmic domain.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Preparation Note

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
